What should the breeding set up be for 2 Leopard Geckos. I read that the substrate should be vermiculite and that it should have a nest box.

At least a 20 gallon long for 2 geckos
Adequate hides so the female can get away from the male if she wants to
As you said, a lay box which could have moss, vermiculite or coco fiber in it (I prefer coco fiber)
All the other stuff you would usually have in a gecko tank.

If you're planning to keep your males and females separate and only put them together briefly for breeding, then you need the lay box in the female's enclosure only.
